The 27-year-old came into the tournament ranked 114th - he's now the first male grand slam debutant to make a semi-final.
His four-sets win over Grigor Dimitrov sets up a meeting with defending champion Novak Djokovic.
Karatsev says his success has centred on self confidence.
Serena Williams has powered past Simona Halep in straight sets to book a last four spot against Naomi Osaka.
